NanoLuc Genetic Reporter Vectors

NanoLuc?（Nluc）luciferase（NanoLuc? 螢光素酶）是一個經(jīng)過基因工程改造的小分子酶（19.1kDa），是性能卓越的生物發(fā)光報告基因。該酶比螢火蟲（Photinus pyralis）或海腎（Renilla reniformis）螢光素酶的發(fā)光亮 100 倍，它使用一種新型底物——furimazine，可產(chǎn)生高強度、輝光型發(fā)光。生物發(fā)光反應(yīng)不依賴 ATP，同時抑制背景發(fā)光以獲得最高檢測靈敏度。

目前已構(gòu)建多種類型的 NanoLuc? 螢光素酶報告基因載體，以滿足不同的實驗需求。非融合的 Nluc 提供了最大的光輸出和靈敏度，NanoLuc?-PEST（NlucP）將蛋白表達水平變化與轉(zhuǎn)錄活性改變密切偶聯(lián)，并增加信噪比，融合 N- 末端分泌信號（secNluc）的 NanoLuc? 螢光素酶適用于分泌型報告基因試驗。使用 Nano-Glo? 螢光素酶檢測試劑，發(fā)光量在跨越 1,000,000 倍的酶濃度范圍內(nèi)呈線性關(guān)系，且信號半衰期≥ 2 小時。

NanoLuc? 螢光素酶具有許多使之成為出色報告基因蛋白的物理特性：
? 分子量非常小，單體酶（171 個氨基酸；513bp）
? 熱穩(wěn)定性高（Tm=60℃）
? 在很寬的pH 范圍內(nèi)有活性（pH 值6-8）
? 沒有翻譯后修飾或二硫鍵
? 均勻分布在細胞中
? 發(fā)射光譜非常適合于生物發(fā)光共振能量轉(zhuǎn)移（BRET；λmax=465nM）。

為滿足利用報告基因研究轉(zhuǎn)錄調(diào)控，我們提供多種表達不同類型 NanoLuc? 螢光素酶（非融合型Nluc、PEST 不穩(wěn)定型NlucP、分泌型secNluc）的質(zhì)粒載體。不同的 pNL 載體用途如下：
? pNL1：克隆已知或預(yù)測的啟動子區(qū)域
? pNL2：克隆已知或預(yù)測的啟動子區(qū)域，并通過潮霉素（Hygromycin）篩選建立一個穩(wěn)定克隆的細胞系
? pNL3：克隆不需要基本啟動子的結(jié)合位點或應(yīng)答原件（如 pNL3.2.NF-κB-RE 載體）
? 提供非融合和分泌型 Nluc 對照質(zhì)粒。pNL 載體系列以pGL4 為基礎(chǔ)設(shè)計，以方便從現(xiàn)有的質(zhì)粒上進行序列轉(zhuǎn)移。載體骨架設(shè)計還去掉了許多轉(zhuǎn)錄因子結(jié)合位點和其他潛在的調(diào)控元件以減少反常結(jié)果的發(fā)生。對Nluc基因突變體進行了密碼子優(yōu)化，并去除了許多潛在調(diào)控元件或其他不必要的功能（如常見的限制性內(nèi)切酶位點）。
