To date, no Z. rouxii episomal plasmid has been available with a multiple cloning site. To construct the first Z. rouxii episomal plasmids, a 1.2-kb pSR1 fragment was selected as a replicon, as this fragment was previously shown to contain ARS and elements for stable partitioning into subsequent generations (Jearnpipatkul et al., 1987a, b). Three Z. rouxii-specific episomal plasmids with multiple cloning sites were constructed, pZEU, pZEA and pZEL, harbouring ScURA3, ZrADE2 or ZrLEU2 as marker genes, respectively, and pSR1 as a replicon
