Medium:
80% RPMI 1640 + 20% h.i. FBS
Subculture:
seed out at ca. 0.8-1.5 x 106 cells/ml in 24-well plates; maintain at ca. 0.6-1.5 x 106 cells/ml; optimal split ratio of 1:2 to 1:3 every 2-5 days; cells grow slowly after recovery from liquid nitrogen
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 60-80 hours
Harvest:
cell harvest of ca. 2.5-3.0 x 106 cells/ml
