BioVector? NCI-H1975/AR cells were established from NCI-H1975 cells through dosage-escalation of OSI from 0.03 μM to 1.5 μM for about 6 months. The cell viabilities of NCI-H1975 and NCI-H1975/AR cells following OSI treatment were studied by 3-(4,5-dimethylthiazol-2-yl)-2, 5-Diphenyltetrazolium bromide (MTT) assay. The cell viability of NCI-H1975/AR cells did not decrease as significantly as that of NCI-H1975 cells after exposure to OSI for 72h. The IC50 values of OSI for NCI-H1975 and NCI-H1975/AR cells were 0.03 μM and 4.77 μM, respectively (Figure 1C). To further confirm the resistant property of NCI-H1975/AR cells to OSI, the colony formation abilities of NCI-H1975 and NCI-H1975/AR cells after treatment with OSI were detected. Treatment of NCI-H1975 cells with 0.03 μM and 0.5 μM OSI decreased the cell colony formation. However, the colony formation of NCI-H1975/AR cells was not decreased after treatment with OSI, even at the concentration of 0.5 μM OSI.
