EB66 cells are expanded to required pre-infection densities in BioVector? EB66Complete Medium-I (Catalog No. BioVector?-931135M1) growth medium. EB66 cellsare normally grown at 37±1oCand 7.5% CO2 in shaker flasks at 120 rpm.
Virusinfection and production is induced with the addition of seed virus and 1.5 -1.8 times (v/v) pre-warmed BioVector? EB66 Complete Medium-II (Catalog No.BioVector?-931135MII) medium to growth medium. Standard infection celldensities are >2 x 106 viable cells/mL (prior to dilution withproduction medium) at a multiplicity of infection (MOI) of 0.01 - 0.10.
Optimalconditions for virus amplification may vary depending on viral construct andprocess conditions. Recommended post-infection conditions are 33 to 37 °C and7.5 % CO2 in shaker flasks at 120 rpm.
Cryopreservation
