The Bucillus subtilis 168 chromosome carries an integrated

plasmid sequence encoding tetracycline

resistance (Tc? but that the expression

of the Tc’ gene appears to be repressed. When

staphylococcal Tc’ plasmid DNA segments

lacking an autonomously replicating property

are introduced into B. subtilis, the integrated

plasmid sequence seems to be excised from

the chromosome resulting in formation of an

active autonomous Tc’ plasmid. The plasmid

has a length of 4.7 kb and was named

pNS198 1. Preliminary data showed that

pNS 198 1 is essentially the same as pBC16,

formerly found in B. cereus by Bernhard et

al. (I). To study the molecular mechanism of

the repression of the Tc’ gene in the integrated

state, analyses of the mode of insertion of the

pNS 198 1 sequence into the chromosome and

studies of the organization of plasmid

pNS 198 1 are necessary. As a first stage in performing

the above, we have derived a restriction

map of pNS1981 for 11 restriction enzymes.

This map has been orientated with

respect to the unique BumHI site and is shown

in Fig. 1.
