■ 產品概述：

Stbl2 Competent Cells have been prepared by apatented modification of the procedure of Hanahan . These cells are suitablefor the cloning of unstable inserts such as retroviral sequences or directrepeats .

For optimal performance, perform expression in S.O.C.Medium and incubation on antibiotic plates at 30°C. Stbl2 Competent Cells are aderivative of JM109. The mcrA mutation and the mcrBC-hsdRMS-mrrdeletion allowfor the cloning of genomic sequences that are methylated. These cells are notcapable of blue/white selection with plasmids containing α-complementationsequences.

■ 產品儲存：

-80°C 保存，避免反復凍融。

■ 使用步驟：

1. 把感受態細胞置于冰中融化。

2. 把100uL的感受態細胞移至滅菌處理的試管內。

3. 加入用于轉化的DNA（10 ng以下）。

4. 冰中放置30分鐘。

5. 42℃放置45秒。

6. 冰中放置2～3分鐘。

7. 加入30℃預溫好的SOC培養基，使終體積為1 ml。

8. 30℃振蕩培養1小時（160～225 rpm）。

9. 取適量涂布瓊脂平板培養基。

10. 30℃過夜培養。

■ 注意事項：

1. 請用低溫運輸。

2. 不立即使用的感受態細胞請在-80℃下保存（融化后的感受態細胞不能再凍結保存）。

3. 轉化時請使用傳熱性能較好的試管。一般的 1.5 ml 微型離心管也可使用，但轉化效率會有所下降（此時請在使用方法5.的42℃下放置60秒）。

4. 對100 uL的感受態細胞，用于轉化的質粒DNA量請控制在10 ng以下，并保證DNA 溶液的體積在20 uL以下。否則會影響轉化效率。

5. 使用SOC培養基的地方也可使用 LB 培養基，但此時轉化效率會有所下降。
