■ 產(chǎn)品概述：

Stbl2 Competent Cells have been prepared by a patented modification of the procedure of Hanahan . These cells are suitable for the cloning of unstable inserts such as retroviral sequences or direct repeats .

For optimal performance, perform expression in S.O.C. Medium and incubation on antibiotic plates at 30°C. Stbl2 Competent Cells are a derivative of JM109. The mcrA mutation and the mcrBC-hsdRMS-mrrdeletion allow for the cloning of genomic sequences that are methylated. These cells are not capable of blue/white selection with plasmids containing α-complementation sequences.

■ 產(chǎn)品儲(chǔ)存：

-80°C 保存，避免反復(fù)凍融。

■ 使用步驟：

1. 把感受態(tài)細(xì)胞置于冰中融化。

2. 把100uL的感受態(tài)細(xì)胞移至滅菌處理的試管內(nèi)。

3. 加入用于轉(zhuǎn)化的DNA（10 ng以下）。

4. 冰中放置30分鐘。

5. 42℃ 放置45秒。

6. 冰中放置2～3分鐘。

7. 加入30℃預(yù)溫好的SOC培養(yǎng)基，使終體積為1 ml。

8. 30℃振蕩培養(yǎng)1小時(shí)（160～225 rpm）。

9. 取適量涂布瓊脂平板培養(yǎng)基。

10. 30℃過(guò)夜培養(yǎng)。

■ 注意事項(xiàng)：

1. 請(qǐng)用低溫運(yùn)輸。

2. 不立即使用的感受態(tài)細(xì)胞請(qǐng)?jiān)?/span>-80℃下保存（融化后的感受態(tài)細(xì)胞不能再凍結(jié)保存）。

3. 轉(zhuǎn)化時(shí)請(qǐng)使用傳熱性能較好的試管。一般的 1.5 ml 微型離心管也可使用，但轉(zhuǎn)化效率會(huì)有所下降（此時(shí)請(qǐng)?jiān)谑褂梅椒?/span>5.的42℃下放置60秒）。

4. 對(duì)100 uL的感受態(tài)細(xì)胞，用于轉(zhuǎn)化的質(zhì)粒DNA量請(qǐng)控制在10 ng以下，并保證DNA 溶液的體積在20 uL以下。否則會(huì)影響轉(zhuǎn)化效率。
