載體簡介
pPIC9K載體中存在卡那抗性基因,允許利用卡那抗性在酵母體內篩選多克隆拷貝,載體其余部分與pPIC9載體完全一樣。pPIC9K載體能夠使用的畢赤酵母宿主菌是KM71和GS115。
pPIC9K載體詳細信息如下:
• pPIC9載體大小9276 bp,是融合表達載體。
• 載體構建過程中,可供使用的單一限制性內切酶位點為SnaB I, EcoR I, Avr II, Not I.
• 利用alpha因子分泌信號肽,分泌表達蛋白基因。
• 在載體構建過程中,你的基因必須保證與信號肽的起始密碼子的讀碼框一致。
• 畢赤酵母中利用HIS4進行篩選。
• 為了將基因插入GS115或KM71的AOX區, 使用SacI限制性內切酶線性化質粒(GS115中產生His+Mut+基因型,KM71中產生His+ MutS基因型)
•為了將基因插入GS115或KM71的His4區, 使用Sal I或者Stu I限制性內切酶線性化質粒(GS115中產生His+ Mut+基因型,KM71中產生His+ MutS基因型)
•將基因插入GS115的AOX1區域,需要使用Bgl II線性化質粒 (產生His+MutS基因型)。
